In Vitro Response of Gynecological Cancer Cell Lines to the GnRH Agonist, Medroxyprogesterone Acetate and Tamoxifen

Abstract

|
There are many evidences that some gynecological tumors are sex steroid hormone dependent and estrogen is known to be a promotor of sex steroid hormone dependent tumor.
The direct antitumoral effects of GnRH agonists on gynecological cancers have been surmised from clinical obsrervations and in vitro studies. Repeated administration of GnRH agonists induced pituitary desensitizatio followed by decrease in
gonadotropin
secretion and estradiol synthesis. Thus reversible hypogonadism is produced. Consequently, estrogen dependent diseases can be ttreated successfully with GnRH analogues.
Progesterone has been used as therapeutic agent against estrogen tumors, because of its antiestrogenic effect. Medroxyprogesterone acetate has been shown to improve life quality and overall ell-being in patients with advanced breast cancer and
uterine
adenocarcinoma.
Tamoxifen, as antiestrogens, have a direct antitumor action and are widely used in cancer therapy.
Using clonogenic assay, we examined the direct growth inhibitory effect of these agents on gynecological cancer cell lines.
We wished to define accurately the relationship between specific tumor receptors quantitation and response to hormone manipulation in a large series in vivo.
